• Antibody is tested in direct ELISA.
• For immunohistochemistry (PFA-fixed tissues): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling for 20 minutes at room temperature.

Target Information

Beta-2-microglobulin (B2M) is a 12–14 kDa protein that forms a subunit of the major histocompatibility complex (MHC) class I. It is associated with HLA Class I antigens and expressed on various cell types including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial and epithelial cells, but absent on erythrocytes.
It is also found in free form in plasma. The amino acid sequence of B2M is homologous to the constant regions of immunoglobulin heavy and light chains, with a predominantly beta-pleated sheet structure.
B2M is involved in antigen processing and presentation, TCR signaling, and downstream signaling in naive CD8+ cells. It is encoded by the B2M gene on chromosome 15. Mutations in this gene may cause hypercatabolic hypoproteinemia. B2M levels can be used to assess renal function and serve as a prognostic marker for HIV infection.
