Product Specific Information
Description:
BM8 monoclonal antibody reacts with mouse F4/80 antigen (~160 kDa surface receptor), belonging to the EGF-TM7 family. It contains seven EGF-like domains, seven transmembrane sequences, and a C-terminal domain homologous to TM7 superfamily members. F4/80 is highly expressed on mature macrophages and serves as a key marker for this population. It can also be expressed on eosinophils, Langerhans cells, and certain dendritic cell subsets.
Applications Reported:
Suitable for flow cytometric analysis.
Applications Tested:
Validated for flow cytometry using mouse peritoneal exudate cells. Recommended ≤1 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize titration per assay.
Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Use a 610/20 band pass filter (PE-Texas Red® equivalent). Ensure instrument compatibility.
Light Sensitivity:
Tandem dye sensitive to photo-induced oxidation. Protect vial and samples from light.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:
- IC Fixation Buffer (Product #00-8222)
- 1-step Fix/Lyse Solution (Product #00-5333)
Minimal impact on brightness and FRET efficiency. Clone-specific performance should be empirically determined.

Target Information
F4/80 antigen is a 160 kDa surface glycoprotein, part of the EGF TM7 family, sharing 68% amino acid identity with human EMR1. Expression varies during macrophage maturation and activation. Present in tissue macrophages (Kupffer cells, Langerhans cells, microglia, gut, peritoneal cavity, lung, thymus, bone marrow stroma, spleen red pulp). Also found in some dendritic cells, absent in T-cell areas of spleen/lymph node. Suggested role in generation of efferent CD8⁺ regulatory T cells.
