Target Information
MELK is a serine/threonine kinase related to the AMPK family, linked to the cell cycle and overexpressed in various cancers. It phosphorylates substrates including BCL2L14, CDC25B, MAP3K5/ASK1, and ZNF622. MELK acts as an activator of apoptosis by phosphorylating MAP3K5/ASK1 and regulates cell cycle progression through CDC25B phosphorylation and localization to centrosome and spindle poles. It plays a key role in cell proliferation and carcinogenesis, essential for embryonic and postnatal neural progenitor proliferation. MELK also inhibits spliceosome assembly during mitosis via ZNF622 phosphorylation, contributing to nuclear redirection and possibly affecting mammary carcinogenesis by modulating BCL2L14 activity.
