Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha)는 골수계 세포(과립구, 수지상세포, 대식세포, 비만세포, 조혈줄기세포 등)에 발현되는 수용체형 막단백질입니다.
이 단백질은 활성화된 여러 티로신 키나아제(EGFR, PDGFR, src, insulin receptor 등)의 기질로 작용하며, 수용체 티로신 키나아제 신호전달 경로의 음성 조절에 관여합니다.
SIRP alpha는 CD47과 결합하여 세포 내 ITIM 영역의 인산화를 유도하고, SHP-1 및 SHP-2 인산화효소를 모집 및 활성화시킵니다.
이 상호작용은 대식세포의 식작용 억제, T 세포 및 수지상세포의 양방향 활성화 등 다양한 면역 조절 과정에 관여합니다.
또한 SIRP alpha는 성장인자, 인슐린, 종양유전자, 세포 부착 등 다양한 자극에 대한 세포 반응 조절에 관여하며, 암세포에서 CD47의 과발현을 통해 대식세포 매개 암세포 제거를 억제하는 역할을 합니다.
