Target Information
This gene encodes a protein that is a subunit of troponin, a regulatory complex located on the thin filament of the sarcomere.
Troponin regulates striated muscle contraction in response to changes in intracellular calcium concentration and is composed of three subunits:
- Troponin C: binds calcium
- Troponin T: binds tropomyosin
- Troponin I: inhibitory subunit
TNNT1 encodes the slow skeletal troponin T subunit.
Mutations in this gene cause nemaline myopathy type 5 (Amish nemaline myopathy), a neuromuscular disorder characterized by muscle weakness and rod-shaped inclusions in skeletal muscle fibers.
Affected infants often die due to respiratory insufficiency, typically within the second year.
Multiple transcript variants encoding different isoforms have been identified for this gene.
