Product Specific Information

The UC10-4B9 monoclonal antibody reacts with mouse CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed at low levels on activated T cells and binds B7 family members CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28. While CD28 acts as a potent costimulator, CTLA-4 inhibits T cell activation by restricting IL-2 secretion and proliferation.

Due to intracellular localization of CTLA-4, complete detection may require assessment of both surface and intracellular expression.

Applications Reported: Flow cytometry, immunoprecipitation, and in vitro functional studies (use Functional Grade purified UC10-4B9, Product #16-1522, for functional assays).

Applications Tested: Tested by flow cytometric analysis of resting and activated mouse splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Optimal titration required per assay.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28 but delivers inhibitory signals, suppressing T cell activation. Intracellular CTLA4 is also found in regulatory T cells, contributing to their suppressive function.
