Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to downstream signaling pathways, playing critical roles in regulating cell death and stress responses.
TRAF proteins share conserved domains enabling receptor and TRAF–TRAF interactions, leading to activation of various signaling cascades.
TRAF2 and TRAF3 bind to CD40, a TNF receptor superfamily member involved in B cell immunity, and are essential for NF-κB activation in mouse B lymphocytes.
TRAF2, together with TRAF6, is necessary for CD40 signaling in non-hematopoietic cells and interacts with lymphotoxin-beta receptor (LTβR) in association with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
