The monoclonal antibody 3D10 recognizes Ly-49H, a member of the C-type lectin Ly-49 multigene family of receptors found on natural killer (NK) cells.
Ly-49H, similar to Ly-49D, lacks the ITIM (immunoreceptor tyrosine-based inhibitory motif) and contains an arginine residue in the transmembrane domain, suggesting it functions as an activation receptor.
Ly-49H is expressed in a subset of NK cells but not in NKT (NK1.1⁺CD3⁺) cells. NK cells can express multiple Ly-49 proteins. Preferential staining of Ly-49H with Ly-49D has been observed. Expression is confirmed in C57BL/6 and NWNA mice, but not in Balb/c or DBA/2 strains. No cross-reactivity with Ly-49A, C, D, or G2 has been detected.
Addition of monoclonal antibody 3D10 to cell cultures induces lysis of target cells.

Applications Tested: Tested on C57Bl/6 spleen cells; usable at ≤1 µg/test (in 100 µL final volume).
Cell numbers can vary from 10⁵–10⁸ cells/test. Optimize antibody titration for best results.
