Target Information

Mitochondrial transcription factor A (TFAM) is a crucial protein encoded by nuclear genes but transported to mitochondria, where it maintains and regulates mitochondrial DNA (mtDNA). It controls mtDNA replication and transcription, ensuring proper mitochondrial function. TFAM contains an LC3 interacting region (LIR) motif that binds autophagy proteins such as LC3, supporting the autolysosomal pathway to remove leaked mtDNA and limit inflammation.

TFAM expression varies across tissues, affecting mitochondrial function. High TFAM levels can repress mtDNA expression in skeletal muscle, leading to oxidative phosphorylation (OXPHOS) deficiencies, while in cardiac tissue, balanced TFAM-to-mtDNA ratios preserve OXPHOS capacity. TFAM is essential for mitochondrial genome packaging, stability, and replication, with disruptions linked to neurodegeneration and other mtDNA-related diseases.
