Target Information

EGFR (Epidermal growth factor receptor, HER1, ErbB1) is encoded by the EGFR gene located on chromosome 7 in humans. EGFR belongs to the HER/ERbB family of proteins that includes three other receptor tyrosine kinases: ERbB2, ERbB3, and ERbB4.
EGFR is a transmembrane receptor; binding of ligands such as EGF and TGF-alpha leads to dimerization and autophosphorylation of tyrosine residues. This activation triggers downstream signaling cascades including Ras–ERK, PI3K–Akt, Jak–STAT, and PKC pathways, promoting cell growth and proliferation.
Phosphorylation at Y1086 enables GRB2 binding and MAPK pathway activation. EGFR mutations are linked to lung cancer, and overexpression is associated with poor prognosis in various tumors including those of the head, neck, brain, bladder, stomach, breast, lung, and others.
