Target Information
Tight junctions are specialized regions of cell-cell contact abundant in luminal epithelial cell sheets. In electron micrographs, they appear as belt-like bands of sealing strands that encircle each cell. These strands form a “molecular gasket” preventing ions, water, and molecules from leaking between cells, maintaining the barrier and polarity of epithelial tissues.
Peripheral membrane proteins such as ZO-1, ZO-2, ZO-3, cingulin, and symplekin are associated with tight junctions and may play roles in assembly, maintenance, and permeability regulation. Actin filaments also contribute to tight junction regulation.
Claudin-1 and Claudin-2 are ~22 kDa transmembrane proteins forming structural components of tight junction strands. Claudin-2 functions as a paracellular channel with cation (Na⁺) selectivity and is primarily expressed in the liver and kidney, with minor expression in the brain.
