Target Information
NANOG (Nanog homeobox) is a divergent homeodomain protein that regulates pluripotency and differentiation of undifferentiated embryonic stem cells. NANOG mRNA is expressed in pluripotent mouse and human cell lines, but absent in differentiated cells. Human NANOG shares 52% overall amino acid identity and 85% identity in the homeodomain with mouse NANOG.
Human NANOG maps to gene locus 12p13.31, while mouse NANOG maps to 6 F2. Murine embryonic NANOG expression is detected in the inner cell mass of the blastocyst. High levels of NANOG expression are found in undifferentiated N-Tera embryonal carcinoma cells. NANOG functions as a transcription regulator involved in inner cell mass and embryonic stem (ES) cell proliferation and self-renewal.
NANOG plays a key role in embryonic development and is useful in stem cell research for regenerative medicine. Induced pluripotent stem (iPS) cells can be generated by expressing POU5F1 (Oct-4), Sox2, Klf4, Lin28, and c-Myc, or by plasmid-based expression of NANOG, Sox2, Klf4, and c-Myc without viral introduction. Overexpression of NANOG promotes S-phase entry and proliferation.
Diseases associated with NANOG dysfunction include teratocarcinoma and germ cell/embryonal cancer.
