Product Specific Information
The pY20 monoclonal antibody recognizes phosphorylated tyrosine residues (p-Tyr) on proteins. Tyrosine phosphorylation is a key mechanism in intracellular signaling cascades. Detection of p-Tyr residues aids in the characterization and purification of phosphorylated proteins and their associated biochemical pathways.
Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by intracellular staining of normal human peripheral blood cells at 5 µL (0.06 µg) per test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.
Fluorophore Information:
eFluor™ 450 is an alternative to Pacific Blue™, excited at 405 nm and emitting at 445 nm. Compatible with Violet laser detectors.

- Protocol A: Two-step for cytoplasmic proteins (surface + intracellular detection)
- Protocol B: One-step for nuclear proteins (transcription factors + phosphorylated intracellular proteins)

Target Information
Tyrosine phosphorylation plays a critical role in mitogenic signal transduction and oncogenic transformation. The fluorescein (FITC) conjugate of clone pY20 is particularly useful for direct detection in immunohistochemical and immunocytochemical assays where secondary antibody use may interfere.
