Target Information

IRS-1, a major substrate of the insulin receptor, is phosphorylated in response to stimulation by insulin, IGF-1, and IL-4. It undergoes phosphorylation on serine, threonine, and tyrosine residues in various tissues. Casein kinase II mediates part of the insulin-stimulated phosphorylation of IRS-1, with Thr502 and Ser99 identified as major phosphorylation sites. IRS-1 also contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with the Ser270 motif located in the phosphotyrosine binding domain responsible for insulin receptor interaction.
