Target Information

Transforming Growth Factor Beta-1 (TGF beta-1, TGFB1) is a cytokine member of the TGF-beta superfamily, widely expressed in tissues. It is stored in the extracellular matrix as a latent complex with its pro-domain. Activation requires binding of αV integrin to an RGD sequence in the prodomain, exerting force to release active TGF beta-1.

Latent forms consist of TGF beta-1, the latency-associated peptide (LAP), and a latent TGF-beta-binding protein (LTBP). TGF beta-1 regulates proliferation, differentiation, adhesion, and migration across various cell types. It modulates other growth factors both positively and negatively. The mature peptide forms homodimers or heterodimers with other TGF beta family members. Mutations in TGFB1 are associated with Camurati-Engelmann disease and cystic fibrosis, and its gene is often upregulated in tumor cells.
