Target Information
NLK is a serine/threonine kinase involved in signal transduction and cellular communication. It acts as a positive effector of the non-canonical Wnt signaling pathway, functioning downstream of WNT5A, MAP3K7/TAK1, and HIPK2. Activation of this pathway leads to phosphorylation of SETDB1, forming the NLK-SETDB1 complex that interacts with PPARG, resulting in histone methylation and transcriptional silencing of PPARG target genes. This process inhibits adipogenesis and promotes osteoblastogenesis in mesenchymal stem cells (MSCs).
NLK also acts as a negative regulator of the canonical Wnt/β-catenin signaling pathway by phosphorylating TCF7L2/TCF4 and LEF1, disrupting their complex with β-catenin and promoting LEF1 degradation. Additionally, NLK negatively regulates the Notch signaling pathway by phosphorylating NOTCH1, preventing the formation of the active transcriptional complex with RBPJ/RBPSUH and MAML1. It also negatively regulates the MYB family of transcription factors.
