Target Information
RUNX2 is a nuclear protein and a member of the RUNX family of transcription factors containing a Runt DNA-binding domain. It is essential for both membranous and endochondral bone formation. RUNX2 regulates osteoblastic differentiation and skeletal morphogenesis and acts as a scaffold for nucleic acids and regulatory factors involved in skeletal gene expression. The protein binds DNA as a monomer or as part of a heterodimeric complex with higher affinity. It also enhances TGFBR1 expression in osteoblasts and cooperates with DLX5 or related factors to activate osteoblast-specific genes. Mutations in RUNX2 are associated with cleidocranial dysplasia (CCD).
