Target Information
GAD-65와 GAD-67은 group II decarboxylase 계열 단백질로, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 반응의 속도 제한 단계를 촉매합니다. 두 단백질 모두 뇌에서 발현되지만, GAD-65는 신경 말단의 시냅스 소포막에 국한되고, GAD-67은 세포 전체에 분포합니다. GAD-67은 기본적인 GABA 합성을 담당하며, GABA 요구량이 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 돕습니다. GAD-65의 결손은 GABA 신경전달에 손상을 주지만, GAD-67의 결손은 치명적입니다. 대체 스플라이싱으로 인해 GAD-67의 두 가지 isoform(GAD-67 주요형과 GAD-25 소형)이 존재합니다.
