• For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
• Mouse anti-Human CD163 antibody, clone EDHu-1 recognizes the human CD163 cell surface antigen, a 130–140 kDa glycoprotein expressed by tissue macrophages.
• CD163 is not expressed by resting peripheral blood leucocytes but expression may be induced on monocytes by culture in dexamethasone.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein and a member of the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and aiding iron recycling through endocytosis of hemoglobin/haptoglobin complexes.

CD163 is expressed exclusively on human monocytes and macrophages, predominantly in the late phase of inflammation. It is present on all circulating monocytes and most tissue macrophages, except those in mantle zones, germinal centers, interdigitating reticulum cells, and Langerhans cells.

CD163 expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding into plasma or supernatant. The receptor binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, with higher affinity for HP1F phenotype complexes. CD163 signaling involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
