Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 세포자멸사(apoptosis), 분화 등 다양한 세포 과정을 조절하는 단백질입니다. SMAD2는 TGF-beta 수용체와 상호작용하여 신호 전달 경로의 핵심 역할을 수행합니다.
ERK1에 의해 MH1 도메인의 calmodulin-binding 영역 내 threonine 8이 인산화되면 SMAD2의 전사 활성화가 촉진되며, 이는 calmodulin에 의해 음성 조절됩니다.
TGF-beta 신호에 반응하여 SMAD2는 수용체에 의해 인산화되고, SARA와의 결합이 해제되며 SMAD4와 결합합니다. SMAD4와의 결합은 SMAD2가 핵으로 이동하여 표적 프로모터에 결합하고 전사 억제 복합체를 형성하는 데 중요합니다.
SMAD2는 activin type 1 receptor kinase에 의해서도 인산화되어 activin 신호를 매개할 수 있습니다. 또한, 대체 스플라이싱된 전사 변이체가 보고되었습니다.
