Product Specific Information

The eBioGHI/61 monoclonal antibody reacts with human CD163 (Haemoglobin Scavenger Receptor, M130), a member of the macrophage scavenger receptor family. CD163 is expressed by tissue macrophages, circulating blood monocytes, and possibly hematopoietic progenitor cells. It functions as a high-affinity scavenger receptor for the hemoglobin-haptoglobin complex, aiding in hemoglobin clearance from the blood.

Upon ligand binding, CD163 initiates signal transduction leading to IL-6 and IL-10 production. IL-6 and IL-10 can induce CD163 expression, while proinflammatory stimuli such as LPS or TNF-α cause shedding of soluble CD163.

Applications Tested: Tested on normal human peripheral blood cells at ≤1 µg/test (100 µL final volume). Optimal titration recommended.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein and receptor for the hemoglobin-haptoglobin complex. It protects tissues from hemoglobin-mediated oxidative damage and may contribute to iron recycling via endocytosis and heme breakdown.

CD163 is expressed on monocytes and macrophages, particularly in late inflammation phases. It is upregulated by IL-10, IL-6, and dexamethasone, and shed by LPS or PMA stimulation. CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F) complexes.
