Target Information
Cyclooxygenase 2 (COX2) catalyzes the conversion of arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. The reaction involves two steps: a cyclooxygenase reaction converting arachidonate to PGG2 and a peroxidase reaction reducing PGG2 to PGH2.
COX2 is constitutively expressed in tissues such as the endothelium, kidney, and brain, and is upregulated under pathological conditions including cancer and inflammation. Elevated COX2 levels are associated with increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis. In cancer cells, COX2 facilitates prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance. COX2 activity is naturally inhibited by calcitriol (active Vitamin D), and glucocorticoids can repress PTGS2 gene activity. COX2 is a target of NSAIDs such as aspirin, which reduce inflammation-related pain and swelling.
