Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. The conversion involves two reactions: a cyclooxygenase (COX) reaction converting arachidonate to prostaglandin G2 (PGG2), and a peroxidase reaction reducing PGG2 to PGH2.
COX2 is constitutively expressed in certain tissues (endothelium, kidney, brain) and upregulated under pathological conditions such as cancer and inflammation. Up-regulation is linked to cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 contributes to prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance. COX2 is naturally inhibited by calcitriol (active Vitamin D form) and repressed by glucocorticoids. It is a target of NSAIDs such as aspirin, which reduce inflammation-related pain and swelling.
