Product Specific Information

PA1-334 detects FXR in mouse samples.
PA1-334 has been successfully used in Western blot procedures.
By Western blot, this antibody detects an ~55 kDa protein representing FXR in mouse liver samples.
An unknown band at ~60 kDa is also observed in western blots.

The PA1-334 immunogen is a synthetic peptide corresponding to residues R(224) K N V K Q H A D Q T V N E D(238) of mouse FXR.
The PA1-334 immunizing peptide (Cat. # PEP-273) is available for use in neutralization and control experiments.


Target Information

FXR (farnesoid X receptor; NR1H4) belongs to the nuclear receptor superfamily of ligand-activated transcription factors (NR1 subfamily) and acts as a receptor for bile acids such as chenodeoxycholic acid, lithocholic acid, and deoxycholic acid.
FXR plays an essential role in bile acid homeostasis through the regulation of genes involved in bile acid synthesis, conjugation, and enterohepatic circulation.
FXR also regulates lipid and glucose homeostasis and is involved in innate immune response.

The FXR-RXR heterodimer binds predominantly to farnesoid X receptor response elements (FXREs) containing two inverted repeats of the consensus sequence 5'-AGGTCA-3', with monomers spaced by 1 nucleotide (IR-1), and also to tandem repeat DR1 sites with lower affinity.
In the liver, FXR activates transcription of the corepressor NR0B2, thereby indirectly inhibiting CYP7A1 and CYP8B1 (involved in bile acid synthesis), implicating histone demethylase KDM1A in epigenomic repression.
FXR activates transcription of SLC27A5/BACS and BAAT (involved in bile acid conjugation), ABCB11/BSEP (bile salt export), ABCC2/MRP2 (secretion of conjugated bile acids), and ABCB4 (phosphatidylcholine secretion in the small intestine).
