Target Information

This gene encodes the coagulation factor XIII A subunit. Coagulation factor XIII is the last zymogen to become activated in the blood coagulation cascade. Plasma factor XIII is a heterotetramer composed of two A subunits and two B subunits. The A subunits have catalytic function, while the B subunits serve as plasma carrier molecules without enzymatic activity. Platelet factor XIII consists only of two A subunits, identical to those of plasma origin.

Upon thrombin cleavage and in the presence of calcium ions, plasma factor XIII dissociates its B subunits to yield the active enzyme factor XIIIa, identical to platelet factor XIII. This enzyme functions as a transglutaminase, catalyzing gamma-glutamyl-epsilon-lysine crosslinking between fibrin molecules, thereby stabilizing the fibrin clot. It also crosslinks alpha-2-plasmin inhibitor or fibronectin to the alpha chains of fibrin.

Factor XIII deficiency is categorized into:

  • Type I deficiency: Lack of both A and B subunits
  • Type II deficiency: Lack of A subunit only

These deficiencies can cause lifelong bleeding tendencies, defective wound healing, and habitual abortion.
