Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, playing key roles in regulating cell death and stress responses. TRAF proteins share a homology region enabling receptor and TRAF–TRAF interactions, activating various signal cascades.
TRAF2 and TRAF3 directly bind CD40, a member of the NF receptor superfamily, and are critical for NF-kappa-B activation in mouse B lymphocytes. TRAF2 with TRAF6 is required for CD40 signaling in nonhemopoietic cells. Additionally, TRAF2 interacts with lymphotoxin-beta receptor (LTbetaR) in association with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
