Product Specific Information

The 15A7 monoclonal antibody reacts with mouse CD95 (Fas, Apo-1), a 40–50 kDa member of the TNFR superfamily. CD95 is expressed by a broad range of hematopoietic and non-hematopoietic cells including monocytes, neutrophils, activated lymphocytes, and fibroblasts. Interaction of CD95 on mature lymphocytes with its ligand (FasL) induces apoptosis and plays an important role in peripheral tolerance.
15A7 reacts with mouse CD95 in BALB/c strain and more weakly in C57Bl/6 strain. It does not induce apoptosis of Fas-expressing cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometry on BALB/c splenocytes and thymocytes. Recommended ≤1 µg/test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. It also exists in a soluble 26 kDa form. Stimulation of FAS leads to aggregation of FADD, formation of the death-inducing signaling complex (DISC), and activation of caspases, triggering apoptosis.
FAS can activate NF-κB, MAPK3/ERK1, MAPK8/JNK pathways, and is involved in proliferative signaling in fibroblasts and T cells. Multiple splice variants exist, some lacking the transmembrane domain and negatively regulating apoptosis. The Fas/FasL system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
