비활성 상태의 proenzyme 형태로 존재하며, proteolytic processing을 통해 활성형 효소로 전환됩니다. Caspase 8은 Fas 및 다양한 세포자멸 유도 자극에 의해 활성화되며, FADD와 상호작용하여 downstream caspases (예: Caspase 9)를 활성화시킵니다.
또한 Caspase 8은 Huntington 병 환자의 뇌 영역 불용성 분획에서 검출되어 신경퇴행성 질환과의 관련성이 제시되었습니다. 과발현 시 세포자멸을 유도하며, ICE 계열 억제제에 의해 차단될 수 있습니다. 다양한 스플라이싱 변이체가 보고되어 있으며, 일부는 완전한 서열이 규명되지 않았습니다.
