Target Information

Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53.
p21 plays a key role in cellular response to genotoxic stress and is a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking cell cycle progression from G1 to S phase or G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 has anti-apoptotic effects by binding and inhibiting caspase 3, ASK1, and JNK.
It exhibits dual roles in carcinogenesis, functioning both as a tumor suppressor and as an oncogene depending on context.
