Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses.
PKR plays a key role in antiviral defense mediated through its activation by double-stranded RNA produced during viral infection. It is involved in IFN-induced innate antiviral response, virus-induced apoptosis, and regulation of cell growth and differentiation.

PKR induces apoptosis by upregulating Fas expression and mediates FADD/Caspase-8 death signaling. Upon viral infection, PKR functions as a dual protein, activating both cell survival and death pathways through kinase-dependent and independent mechanisms.

Additionally, PKR regulates multiple signaling pathways including NF-kappaB, p53, p38, and PDGF. It has been implicated in tumor suppression and malignancy.
Viruses can evade PKR antiviral effects via mechanisms such as inhibition by influenza virus NS1 protein. PKR is also associated with neurodegenerative diseases including Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis.
