Product Specific Information

The 1E7.2 antibody reacts with human and mouse ZAP-70, a cytosolic protein tyrosine kinase (PTK) in the Syk family. It is expressed in T and NK cells and is essential for TCR signaling and development. ZAP-70 binds to phosphorylated ITAMs on TCR complex subunits and becomes phosphorylated by autophosphorylation and by Lck kinase. Phosphorylation enhances kinase activity and downstream signaling. Mutations in ZAP-70 can lead to Severe Combined Immunodeficiency Syndrome (SCID).
The 1E7.2 clone was generated against a KLH-peptide corresponding to residues 282–307 of human ZAP-70. ZAP-70 expression correlates with mutated IgVH in B-CLL, serving as a prognostic marker measurable by intracellular flow cytometry.

  • Tested on human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).
  • Recommended ≤ 0.5 µg/test (100 µL final volume).
  • Cell number: 10⁵–10⁸ cells/test (empirical determination required).
  • Titrate for optimal performance.

Target Information

ZAP-70 is a cytosolic tyrosine kinase of the Syk family, expressed in T-cells and NK cells. It is required for TCR activation. Upon TCR engagement, ZAP-70 is phosphorylated at tyrosines 292, 315, and 319.

  • Tyrosine 315 (Vav binding site) and 319 (Lck binding site) enhance signaling.
  • Tyrosine 292 attenuates activation and signaling.
