Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is detectable during active phases of the cell cycle (G1, S, G2, and mitosis) but absent in resting cells (G0). It localizes to the nucleus during interphase and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.

This antibody also recognizes canine Ki-67 and is validated for intracellular staining followed by flow cytometric analysis. It has been tested using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended usage is 5 µL (0.25 µg) per test, defined as the amount to stain a cell sample in 100 µL volume. Applicable cell counts range from 10⁵ to 10⁸ cells/test.

Brilliant Violet™ 421 (BV421) emits at 423 nm and is suitable for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility before use.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin through its FHA domain and shows cell cycle-dependent localization. Ki-67 is a well-established proliferation marker, absent in G0 cells but increasing through mitosis. It is used to determine the proliferating fraction in neoplasms, correlating with tumor grade and prognosis. Commonly applied as a neuronal marker for cell cycling and proliferation.
