Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive processes such as pregnancy establishment and maintenance.
PR exists as two isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), produced by alternative translation initiation. PR-B is transcriptionally active, regulating genes for endometrial maintenance and inhibition of ovulation. PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Progesterone binding induces conformational changes, dimerization, and binding to progesterone response elements (PRE), resulting in activated transcription. Null mutations in the PGR gene cause reproductive abnormalities.
