Target Information
RNA polymerase (Pol) III synthesizes tRNA, 5S rRNA, 7SL RNA, and U6 snRNA and is overexpressed in many transformed cell lines and tumors. Pol III transcription requires TFIIIB and TFIIIC2 complexes, which include subunits such as TFIIIC220, TFIIIC110, TFIIIC102, TFIIIC90, and TFIIIC63. These are upregulated in malignant cells like ovarian carcinomas. TFIIIC2 recruits RNA Pol III and TFIIIB to promoter elements, potentially contributing to deregulation in cancer. TFIIIB includes TBP, BRF1, and other factors whose expression increases in transformed cells. Tumor suppressors RB and p53, when inactivated, fail to bind and regulate TFIIIB, further promoting malignancy.
