Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon activation, p53 induces transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 has been linked to tumor formation. p53 functions as a key anticancer agent by blocking proliferation of damaged cells. It is expressed at low levels in normal cells but upregulated in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene lead to loss of tumor suppressor activity and are common in various human cancers, including hereditary Li-Fraumeni syndrome.
