Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. p53 mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide deprivation. It up-regulates stress response genes such as p21waf1, contributing to G1 or G2/M arrest.

p53 structure includes:

• N-terminal transactivation domain
• Central DNA-binding domain
• Oligomerisation domain
• C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 plays roles in DNA damage response, apoptosis, aging, oxidative stress, and learning and memory regulation. Mutations in p53, especially in the TP53 gene, are common in human cancers and lead to loss of tumor suppressor function. Germline mutations can cause hereditary cancer syndromes such as Li-Fraumeni syndrome.
