Target Information

The maf oncogene was identified by structural analysis of the AS42 avian transforming retrovirus genome. The Maf family is divided into two subclasses: large Mafs (vMaf, cMaf, MafB, Nrl) and small Mafs (MafF, MafK, MafG). Both subclasses contain leucine zipper motifs enabling homo- and heterodimerization with various bZip transcription factors. Large Mafs possess an acidic transactivation domain absent in small Maf proteins.

Small Mafs can act as positive transcriptional regulators by targeting active dimerization partners to specific DNA elements or as negative regulators by recruiting repressors or forming inactive homodimers. Human MafF, isolated from a human myometrium cDNA library, encodes a 164-amino-acid protein with an extended leucine zipper and no N-terminal transactivation domain. These small Maf proteins are implicated in development, differentiation, hematopoiesis, and stress response, each regulating stress via distinct mechanisms.
