Target Information

Myelin Protein Zero (MPZ), also known as Zero, is a Type I integral membrane glycoprotein that mediates adhesion of spiraling wraps of the myelin sheath, ensuring stable synaptic transmission.
It constitutes approximately 50% of the total protein in the myelin sheath, contributing to structural integrity of peripheral myelin.
MPZ guides compact myelin wrapping through glycine zipper–dependent dimer and tetramer formation. Mutations such as G134R can disrupt multimerization, leading to demyelinating neuropathies including Charcot-Marie-Tooth disease.
The cytoplasmic domain undergoes serine and tyrosine phosphorylation, particularly during peak nerve myelination. MPZ transcript levels are moderate in the brain, abundant in the thymus, and highest in CNS white matter.
