Product Specific Information
Qdot™ Antibody conjugates provide bright fluorescence suitable for detecting low-abundance extracellular proteins. They are similar in size to R-phycoerythrin (R-PE) and compatible with existing fluorophore conjugates. Best excited by UV or violet light, Qdot conjugates have narrow emission profiles and long Stokes shifts, enabling efficient multicolor assays using a 405 nm laser.
Staining:
Use standard buffers such as PBS with 1% BSA. Analyze cells within 18 hours of staining. Dilute only what is needed for one day. Compatible with reagents such as Cal-Lyse™ Lysing Solution and FIX & PERM™ reagents. Some BD FACS™ Lysing Solution batches may interfere with fluorescence.
Instrument Setup:
Optimal excitation at 405 nm (also excitable by 488 nm). Recommended filter: 655/20 nm. Ensure cytometer filters capture emission maxima for accurate analysis.
Storage:
Store at 2–8°C in the dark. Do not freeze. Expires six months from receipt unless otherwise indicated. Photostable and resistant to light degradation. Contains cadmium and selenium; dispose per local regulations.
Target Information
CD4 antigen participates in MHC class II recognition and acts as a co-receptor for HIV. Expressed mainly in T helper lymphocytes, also found in monocytes, macrophages, dendritic cells, thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues. CD4 aids in T-cell activation via association with TCR and Lck kinase and may mediate neuronal damage in immune-related CNS diseases.
