Qdot™ Antibody (Ab) conjugates possess bright fluorescence emission suitable for detecting low-abundance extracellular proteins. They are similar in size to R-phycoerythrin (R-PE) and compatible with organic fluorophore conjugates. Qdot™ Ab conjugates can be excited by any wavelength below their emission maximum, optimally by UV or violet light. The narrow, symmetric emission profiles allow minimal compensation and efficient multicolor assays using a 405 nm violet laser.

• Staining: Use standard buffers such as PBS with 1% BSA. Analyze cells within 18 hours of staining. Dilute only the amount needed for one day.
• Compatibility: Can be mixed with other antibodies; suitable for use with Cal-Lyse™ Lysing Solution and FIX & PERM™ reagents. Note: Some BD FACS™ Lysing Solution batches may interfere with fluorescence.
• Testing: Each lot tested by flow cytometry using human peripheral blood leukocytes.
• Isotype Control: Mouse IgG2a, Cat. No. Q10014.

Instrument Setup

• Optimal excitation with UV or 405 nm light (also compatible with 488 nm laser).
• Ensure the cytometer has appropriate emission filters; for Cat. No. Q10013, use 605/20 nm filter.
• Check for emission overlap in other channels when using multiple lasers.

Storage and Handling

Store reagents at 2–8°C in the dark. Do not freeze. Product expires six months from receipt unless otherwise indicated. Qdot Ab conjugates are photostable but minimize light exposure when used with conventional fluorochromes. Contains cadmium and selenium; dispose of according to local regulations. Refer to SDS for details.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide (LPS) receptor and is released in both secreted and cleaved forms. CD14 interacts with LBP to bind LPS, contributing to immune responses and inflammation. Soluble sCD14 discriminates structural LPS differences and aids in neutralization of foreign LPS. It also binds apoptotic cells and influences allergic, inflammatory, and infectious processes. Alternative splicing produces multiple transcript variants encoding the same isoform. CD14 dysfunction is associated with infections such as Mycobacterium chelonae and Croup.
