Target Information
Protein p21 is a tumor suppressor protein and an inhibitor of Cyclin-dependent kinases (CDK), transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and acts as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking cell cycle progression from G1 to S phase or G2 to M phase after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic activity by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK. p21 thus has dual roles in carcinogenesis, acting both as a tumor suppressor and as an oncogene depending on context.
