Product Specific Information

This 3ZBR4S monoclonal antibody recognizes human and mouse CD247 (CD3z) phosphorylated on Tyrosine 142.
CD3 is a multi-protein complex expressed on thymocytes and T cells, essential for T cell receptor (TCR) signaling.
The CD3 zeta chain initiates the signaling cascade following TCR engagement and contains three ITAM motifs phosphorylated by Src kinases such as Lck.
Phosphorylated ITAMs recruit ZAP-70, amplifying TCR signaling.
Loss or mutation of CD3 zeta impairs T cell development and function.
Phosphorylation occurs early in the TCR signaling cascade, downstream of Lck and upstream of ZAP-70, LAT, and SLP-76.

Recommended Usage: 5 µL (0.25 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test)

Target Information

The CD3z complex transduces antigen-recognition signals into T cells and regulates TCR surface expression.
CD3 subunits (gamma, delta, epsilon, zeta) are immunoglobulin superfamily members encoded on chromosome 11.
They contain cytoplasmic tails with tyrosine-based motifs mediating signal transduction.
CD3 antigen is present on most T lymphocytes and thymocytes but absent on B or NK cells.
Associated diseases include Immunodeficiency 25 and T-B+ Severe Combined Immunodeficiency due to CD3delta/CD3epsilon/CD3zeta defects.
