This 6B10.2 monoclonal antibody reacts with human CD247 (CD3 zeta chain). The CD3 complex consists of gamma, delta, epsilon, and zeta chains that form part of the TCR/CD3 complex. CD3 zeta is expressed on immature and mature thymocytes, T lymphocytes, and NK cells. Signal transduction during T cell activation occurs through tyrosine phosphorylation of ITAMs on the zeta chain.

The 6B10.2 clone recognizes a transmembrane epitope of CD3 zeta, requiring intracellular staining for flow cytometric analysis. It recognizes the zeta chain as a homodimer but not as a monomer.

Target Information

The CD3z complex is essential for transducing antigen recognition signals in T cells and regulating surface expression of the TCR complex. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. They associate with cytoplasmic signaling molecules via tyrosine-based motifs.

CD3 antigen is found on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

It is not expressed on B or NK cells.
Altered CD3 expression is associated with certain immunodeficiencies such as Immunodeficiency 25 and T-B+ Severe Combined Immunodeficiency.
