Survivin (IAP4) is the smallest member of the Inhibitors of Apoptosis Protein (IAP) family. IAPs are involved in cell signaling, division, and metabolism, and show differential expression in nearly all human cancers but not in most normal tissues. Survivin contains only a single BIR domain and lacks a C-terminal RING finger domain. Expression is high during fetal development and in tumors but low in adult tissues. Survivin is expressed in the G2/M phase of the cell cycle and associates with mitotic spindle microtubules. Disruption of this interaction increases caspase-3 activity and induces apoptosis during mitosis. It is also expressed in brain tissues after traumatic injury and may regulate neural cell proliferation. Multiple transcript variants exist, with at least three full-length isoforms identified. Overexpression of survivin in cancer may allow cells to evade apoptotic checkpoints and progress abnormally through mitosis.
