Target Information
Calcineurin binding protein (Cabin-1) and its rat homolog Cain are widely expressed nuclear phosphoproteins that regulate the serine/threonine phosphatase activity of calcineurin, influencing T cell signaling and apoptosis.
Calcineurin is required for the transcriptional activation of cytokines and transcription factors such as NFAT, NFκB, and AP-1 involved in TCR-mediated signaling.
TCR activation induces PKC-mediated hyperphosphorylation of Cabin-1, promoting high-affinity binding to calcineurin, which inhibits calcineurin activity and attenuates TCR signaling.
Cabin-1 also associates with MEF-2 transcription factors, inhibiting MEF-2-mediated gene transcription and apoptosis.
