Target Information
Calcineurin binding protein (Cabin-1) and its rat homolog Cain are widely expressed nuclear phosphoproteins that regulate the serine/threonine phosphatase activity of calcineurin and influence T cell signaling and apoptosis.
Calcineurin is required for cytokine transcriptional activation and activation of transcription factors such as NFAT, NFκB, and AP-1 involved in T cell receptor (TCR)-mediated signaling. Regulation of calcineurin depends on intracellular calcium concentration changes and protein kinase C (PKC) activity.
Upon TCR activation, PKC induces hyperphosphorylation of Cabin-1, enabling high-affinity binding to calcineurin. This complex inhibits calcineurin activity and attenuates TCR-mediated signaling. Cabin-1 also directly associates with MEF-2 proteins, transcription factors regulating apoptosis signaling in T cells, leading to inhibition of MEF-2-mediated gene transcription and apoptosis.
