Product Specific Information

The monoclonal antibody eBio440c recognizes Siglec-H, a protein exclusively found on plasmacytoid dendritic cells (pDC) or type I IFN-producing cells (IPC) in the naive mouse.
Mouse IPC are typically PDCA+, CD11c+, CD11b-, B220+, and Ly-6C+, and rapidly respond to viral infection.

Siglec-H is a transmembrane protein of the Ig superfamily, similar to CD33, that binds sialic acid but lacks the cytoplasmic ITIM domain (immunoreceptor tyrosine-based inhibitory motif). Siglec-H associates with DAP12 for signal transduction.
The eBio440c antibody has been shown to inhibit pDC function by reducing IFNα secretion in response to CpG.

Some mouse strains (e.g., SJL) exhibit higher percentages of pDCs compared to C57BL/6.

A test is defined as the amount of antibody required to stain a cell sample in a final volume of 100 µL. Cell number may vary from 10⁵ to 10⁸ cells/test. Optimal titration is recommended for best performance.
