Target Information

DNA-directed RNA polymerase II subunit RPB1 (POLR2A) is a DNA-dependent RNA polymerase that catalyzes transcription of DNA into RNA using ribonucleoside triphosphates as substrates.
POLR2A is the largest subunit and a catalytic component of RNA polymerase II, responsible for synthesizing mRNA precursors and various non-coding RNAs. It forms the polymerase active center with the second largest subunit.
Pol II serves as the core of the basal transcription machinery, composed of multiple mobile elements that move relative to each other. RPB1 contributes to the core structure, forming the central cleft, clamp, and jaws that engage the DNA template.
During transcription initiation, the DNA template strand is positioned within the active site cleft of Pol II. A bridging helix from RPB1 promotes translocation of Pol II by alternating between straight and bent conformations, facilitating RNA-DNA hybrid movement.
Elongation is modulated by phosphorylation of the C-terminal domain (CTD) of RPB1, which acts as a platform for transcriptional regulators involved in initiation, elongation, termination, and mRNA processing.
Gene expression regulation depends on the balance between methylation and acetylation of CTD lysine residues.
