Description:
The MMA monoclonal antibody reacts with human CD15 (Lewis X). This 3-fucosyl-N-acetyllactosamine carbohydrate moiety is expressed by granulocytes. Monocytes express this structure to varying degrees, while lymphocytes are negative.

Applications Reported:
This MMA antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm. Ensure the instrument can detect this fluorochrome.

Target Information

CD15 (Lewis X, Le(x); stage specific embryonic antigen-1, SSEA-1):
A trisaccharide determinant (3-fucosyl-N-acetyllactosamine) expressed on glycolipids, glycoproteins, and proteoglycans of various cell types such as granulocytes, mast cells, monocytes, macrophages, gastric mucosa cells, nervous system cells, and tumor cells.
Variants include sialyl-Lewis X and sulphated Lewis X. High surface expression of Le(x) promotes calcium-dependent self-aggregation, relevant in embryo compaction and teratocarcinoma cell aggregation. Sialyl-Le(x) and sialyl-Le(a) act as selectin ligands.
CD15 expression is used for the diagnosis of Hodgkin’s disease.
