Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly fused to recombinant proteins at either the N- or C-terminus to enable purification via immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of recombinant proteins for which no specific antibody exists. His-tagged proteins are purified using metal chelate chromatography due to the strong and selective binding of histidine clusters to metal ions such as Ni²⁺.
His tag-specific antibodies facilitate detection or co-immunoprecipitation of tagged proteins.
